氮烯乙茶在离体大鼠肝脏灌流中的生物转化

STUDIES ON BIOTRANSFORMATION OF ETHEOFAZINE IN ISOLATED PERFUSED RAT LIVER

  • 摘要: 用离体大鼠肝脏灌流方法,研究了抗癌新药氮烯乙茶(EDMTP)的生物转化过程。经HPLC和TLC分离纯化及光谱分析,鉴定了两个主要的代谢产物,EDMTP-Ⅰ为未经转化的原型药物,EDMTP-Ⅱ的结构为7-乙基-8-氨基茶碱。另外在静注EDMTP的小鼠血中也分离到EDMTP-Ⅱ,似可说明大鼠与小鼠对EDMTP生物转化的种属差异较小。

     

    Abstract: The biotransformation of etheofazine (EDMTP), a new anticancerdrug, was studied by using isolated perfused rat liver. Two main metabolites were sepa-rated from the perfusate by HPLC and TLC and their chemical structures were determinedby MS,1H-NMR and IR. EDMTP-I is the parent compound. EDMTP-II is7-ethyl-8-aminotheophylline. EDMTP-II was also separated and identified from theblood of mice atter iv of 10 mg/kg. Species difference of biotransformation of EDMTP be-tween rat and mouse seems to be not significant in this study.
