用多中心合成方法快速、同步合成多肽

SIMULTANEOUS MULTIPLE PEPTIDE SYNTHESIS USING THE MULTIPIN METHOD

  • 摘要: 多中心同步多肽合成技术是新药研究组合化学策略的几种主要技术之一。本文报道了用多中心同步多肽合成仪合成十九个长短不一的多肽,其中最长为二十一肽,最短为十二肽。HPLC结果表明,合成粗品肽的纯度在50%~70%之间,电子喷雾质谱支持了序列的准确性。酶联免疫吸附实验结果表明合成粗品肽即可用作新药先导化合物的组合化学研究。

     

    Abstract: In an effort to employ high throughput peptide synthesis technology for bioactive protein research, a pilot study was conducted using the multipin system provided by Chiron Mimotopes.Nineteen peptides were successfully synthesized simultaneously on the pins. All the peptides obtained were of high purity as shown by HPLC and correct sequences as inferred by MS. We concluded that the multipin method is a powerful technique for protein and peptein and peptide research and highly recommend the use of the multipin system as an economic alternative to expensive peptide synthesizer.
