高效液相色谱法测定大鼠血浆中丹参素和原儿茶醛

HPLC DETERMINATION OF DANSHENSU AND PROTOCATECHUIC ALDEHYDE IN RAT PLASMA

  • 摘要: 目的:为了解大鼠im丹参注射液和ig丹参提取物后丹参素和原儿茶醛的吸收情况,建立了高效液相色谱法测定大鼠血浆中丹参素和原儿茶醛的含量。方法:用高效液相色谱内标法测定。色谱柱为ODS柱,流动相为乙腈—0.01 mol.L-1磷酸二氢钾溶液(8∶92),磷酸调至pH 2.8,在280 nm波长处检测。以对羟基苯甲酸为内标。结果:用所建立的方法测定了大鼠im丹参注射液后丹参素和原儿茶醛的血药浓度,并考察了大鼠ig丹参提取物后的吸收情况。结果表明,丹参素和原儿茶醛可经肌肉和胃肠道吸收入血。结论:丹参素和原儿茶醛可作为丹参注射液和含丹参口服制剂的指标成分。

     

    Abstract: AIM: To investigate the absorption of Danshensu (Dhpl) and protocatechuic aldehyde (Pal) after im injection of Salvia miltiorrhiza and ig Danshen extract. METHODS: An HPLC method was developed for determination of Dhpl and Pal in rat plasma by internal standard method. In this system, the mobile phase consists of acetonitrile and 0.01 mol.L-1 KH2PO4 (8∶92, adjusted to pH 2.8 with phosphoric acid). The flow rate was 1.0 mL.min-1, the UV detector was set at 280 nm. 4-Hydroxy-benzoic acid was used as the internal standard. The concentration of Dhpl and Pal in plasma was determined by the new method after im Salvia miltiorrhiza injection and ig Danshen extract. RESULTS: The result showed that Dhpl and Pal can be absorbed into blood after injection and oral administration. CONCLUSION: Dhpl and Pal can be used as the index components of the preparations containing Danshen.
