四氢巴马汀等单胺排空作用的比较

COMPARISON OF TETRAHYDROPALMATINE, TETRABENAZINE AND RESERPINE IN MONOAMINE DEPLETING ACTION

  • 摘要: 利血平(1.5mg/kg,ip)单独给药后24h能显著降低大鼠脑内NA(—60%)和DA(—75%)。预先给予dl-THP(60mg/kg)能使脑内NA和DA进一步下降,分别为正常对照组的15和5%,与单给利血平组有显著性差别。dl-THP同样亦能协同利血平排空5HT的作用。相反,预先给予丁苯那嗪(50mg/kg)则部分拮抗利血平的排空作用。此外,dl-THP与利血平不同,对Na+K+-ATP酶和Mg2+-ATP酶活性无显著影响。l-或d-THP在10-4mol/L的较高浓度下能诱发豚鼠输精管节律性收缩,并不被多种受体阻断剂拮抗。在体研究表明,dl-THP在较大剂量下与利血平相似,降低犬鼠体温。

     

    Abstract: dl-Tetrahydropalmatine (dl-THP), a tetrahydroisoquinoline alkaloid, was found to lower monoamines and increase their acidic metabolites in rat brain and considered to be a short acting monoamine depletor. In the present studies, dl-THP (60mg/kg), in combination with reserpine (1.5mg/kg), caused a further significant decrease in rat brain levels of NA and DA to 15% and 5% of the normal values respectively, compared to 40% and 25%, respectively, after reserpine alone, dl-THP also potentiated the action of reserpine in depleting scrotonin. On the contrary, pretreatment with tetrabenazine (50 mg/kg) partially antagonized reserpine in decreasing the brain levels of monoamines. These results imply that the site of action of THP on the storage mechanism of synaptic vesicle seems to distinguish from that of tetrabenazine though both drugs are short-term monoamine depletors. In addition, dl-THP showed no effect on Na+K+-ATPase and Mg2+-ATPase activities. In this aspect, THP is different from reserpine, dl-THP at the concentration of 10-4 mol/L was found to evoke rhythmic contraction of isolated guinea-pig vas deference and not antagonized by phentolamine, propanolol, haloperidol, atropine or cyproheptadine. In vivo, dl-THP, like reserpine, at relatively large dose lowered the body temperature of rats.
