三尖杉酯碱抑制蛋白质生物合成机制的初步研究

PRELIMINARY STUDIES ON MECHANISM OF INHIBITION OF PROTEIN SYNTHESIS BY HARRINGTONINE

  • 摘要: 本文研究了三尖杉酯碱对珠蛋白的合成、合成珠蛋白肽链的完整性及肽酰—嘌呤霉素结合反应的影响。观察到:在该药作用下,珠蛋白的合成在延迟了几分钟 后才开始受到抑制;所合成的珠蛋白有完整肽链;肽酰—嘌岭霉素结合物的形成不受抑制。这些结果表明,三尖杉酯碱的作用环节可确定在第一个肽键形成之前(包括该环节),这为进一步研究打下了基础。

     

    Abstract: It has been reported that harringtonine inhibits protein synthesis in ceil-free system. In order to study the mechanism of action of harringtonine on protein synthesis, effect of the drug on the initiation and completion of globin synthesis in rabbit reticulocyte lysate and formation of peptidyl-3H-puromycin on rat liver polyribosome were investigated.The results showed that: (1) there was a delay of several minutes in the onset of inhibition following addition of harringtonine to the lysates; (2) complete 3H-globin chains synthesized after the addition of harringtonine were identical to those synthesized in the absence of the drug representing peptides that have passed the initial stages of synthesis when harringtonine was added; (3) formation of peptidylpuromycin complex was not inhibited by harringtonine. These results suggest that harringtonine inhibits protein synthesis at one or more steps prior to the formation of the first peptide bond.
