极限稀释法微孔池培养测定细胞对抗癌药物的剂量反应曲线

THE RESPONSE OF CELLS TO ANTI-TUMOUR DRUGS ESTIMATED BY LIMITING DILUTION IN MICROWELL CULTURE

  • 摘要: 在应用极限稀释法微孔池培养人T-淋巴细胞和K-562白血病细胞系克隆分析法的基础上,将其应用于抗肿瘤药物的敏感性分析,从而建立了一种简便、敏感而又精确的体外测定药物敏感性的方法,其精确度可达10-5~10-6水平。与常规的活细胞计数方法和3H-TdR参入抑制测定法比较,本法较上二法敏感而精确,结果反映了对肿瘤干细胞的杀灭作用。

     

    Abstract: Based on the development of a cloning assay for humanT-lymphocytes and leukaemic cell lines at limiting dilution in microculture, we extended the assay system to include the assessment of sensitivity to antitumour agent.A simple, sensitive and accurate technique has been established using this system. The accuracy can be calculated in the range of approximately 10-5 to 10-6. Comparing the estimation of viable cell count and tritiated thymidine incorporation, this assay appears to be more sensitive and the result reflects the effect of killing tumour stem cells.
