Identification of hydroxylate metabolites of daidzein and its sulfate conjugates in rat urine by LC-ESI/MSn

  • 摘要: 目的鉴定大豆黄素在大鼠体内的羟基化及其结合形代谢产物。方法SD大鼠分别单剂量给药500 mg·kg-1,收集0~24 h尿样。尿样经SPE ODS C18固相萃取柱纯化后,用LC-ESI/MSn对尿样中的代谢物分别进行选择离子监测(SIM)和多级质谱(MSn)分析。结果在大鼠尿中检测到几种尚未在国内外报道过的羟基化代谢产物及其硫酸酯轭合物。结论LC-ESI/MSn法可以快速、简捷、准确地鉴定大豆黄素在大鼠体内羟基化及其结合形代谢产物。

     

    Abstract: AimTo identify the hydroxylate metabolites and its sulfate conjugates of daidzein in rat urine. MethodsUrine samples from 0-24 h were collected after single ig dose of 500 mg·kg-1 daidzein to each of six rats. The urine samples were purified by SPE column (SPE C18) and analyzed with liquid chromatographic-tandem electrospray ionization ion trap mass spectrometry (LC-ESI/MSn) for potential metabolites. ResultsSeveral new hydroxylate metabolites and its sulfate conjugates were found and identified in rat urine. ConclusionLC-ESI/MSn is proved to be a simple, rapid, sensitive and specific technique for identification of the hydroxylate metabolites and its sulfate conjugates of daidzein in rat urine.
