芍药苷增高小鼠恶唑酮结肠炎黏膜中β-防御素表达并减轻病变
Paeoniflorin increases β-defensin expression and attenuates lesion in the colonic mucosa from mice with oxazolone-induced colitis

观察芍药苷对恶唑酮诱导的实验性结肠炎 (IBD) 小鼠的结肠组织黏膜中β-防御素-2 (HBD-2)、IL-6和IL-10表达的影响。通过建立BALB/c小鼠恶唑酮结肠炎模型, 并将其分为正常对照组、恶唑酮诱导组、5-氨基水杨酸 (5-ASA) 组、芍药苷 (20、10及5 mg·kg−1) 治疗组; 进行疾病活动指数 (DAI) 和组织学评分 (HGC); 使用免疫组织化学、实时荧光定量逆转录聚合酶链反应 (RT-PCR) 检测结肠黏膜中HBD-2和细胞因子IL-10、IL-6蛋白及其mRNA的表达。结果显示, 恶唑酮诱导组的DAI和HGC显著高于正常对照组 (P < 0.01), 芍药苷各组和5-ASA组的评分均显著低于恶唑酮诱导组 (P < 0.05, P < 0.01); 恶唑酮诱导组的HBD-2和IL-6表达较正常对照组均显著升高, 且与恶唑酮结肠炎症DAI和HGC呈正相关 (Pearson r = 0.728 (DAI/HBD-2), Pearson r = 0.758 (DAI/IL-6), P < 0.01, respectively; Pearson r = 0.819 (HGC/HBD-2), Pearson r = 0.825 (HGC/IL-6), P < 0.01, respectively); 恶唑酮诱导组的IL-10表达较正常对照组显著降低 (P < 0.01), 且与恶唑酮结肠炎症DAI和HGC呈负相关 (Pearson r = −0.789 (DAI/IL-10), Pearson r = −0.725 (HGC/IL-10), P < 0.01, respectively); 芍药苷各组和5-ASA组均能显著下调恶唑酮诱导组结肠黏膜部位HBD-2、IL-6含量和上调IL-10蛋白和mRNA的表达。结果表明, 芍药苷通过调节HBD-2、IL-6和IL-10的表达来减轻或改善恶唑酮结肠炎小鼠的症状。
Abstract:Previous studies have demonstrated that the Chinese medicine paeoniflorin, derived from the Ranunculaceae plant peony, peony, purple peony root, was able to have anti-inflammatory, anti-ulcer, anti- hypersusceptibility and anti-oxidation activity. In order to elucidate the pesticide effect and the mechanisms by which paeoniflorin exerts its effect of anti-inflammation and immunoregulation on oxazolone-induced colitic mice, disease activity index (DAI) and histological grading of colitis (HGC) were evaluated in animal model. Moreover, the expressions of HBD-2, IL-6 and IL-10 of mice with experimental colitis were observed with immunohistochemistry and RT-PCR in this study. Results showed that DAI and HGC of oxazolone control group was significantly higher than that of normal control group, and that paeoniflorin groups and 5-ASA group, compared with oxazolone control group, could alleviate the symptoms and histological damages of colitic mice (P < 0.05, P < 0.01). The expression of HBD-2 and IL-6 cytokine on the colon of colitic mice was higher than that of normal control, paeoniflorin and 5-ASA groups (P < 0.05, P < 0.01), but the expression of IL-10 is lower than that of normal control, paeoniflorin and 5-ASA groups (P < 0.05, P < 0.01). The positive correlations were demonstrated between the expression of (HBD-2 and IL-6) and DAI (Pearson r = 0.728, Pearson r = 0.758, P < 0.01, respectively), (HBD-2 and IL-6) and HGC (Pearson r = 0.819, Pearson r = 0.825, P < 0.01, respectively), whereas, the negative correlations were demonstrated between the expression of IL-10 and DAI (Pearson r = −0.789, P < 0.01), IL-10 and HGC (Pearson r = −0.725, P < 0.01). It can be concluded that to some extent paeoniflorin effectively alleviate the symptoms of oxazolone-induced colitis through regulating the expression of HBD-2, IL-6 and IL-10.
