四种滋阴药对四种高血压模型的作用

HYPOTENSIVE ACTION OF 4 “YING TONICS”ON 4 HYPERTENSIVE MODELS

  • 摘要: 高血压病的治疗,根据中医辨证论治,多用滋阴药。本文用大白鼠制备4种高血压模型(肾上腺左侧烫伤、睾丸切除、雌鼠每天皮下注射丙酸睾丸酮5毫克共16天及雄鼠每天皮下注射己烯雌酚1—2毫克共16天),并试验4种滋阴药(黄蘗、知母、生地黄及女贞子)对其血压的影响,以尾容器测量大白鼠的血压。睾丸切除、丙酸睾丸酮及己烯雌酚等3型的大白鼠在手术或给性激素后第4周的血压平均值均较手术前非常显著地升高,其中升高20毫米汞柱以上的分别有38,71及88%。肾上腺烫伤型不升压,可能是因烫伤部位太小或太浅,或因未切除肾脏所致。在手术或注射性激素后第6周开始(睾丸切除型于第5周开始),每种模型的大白鼠各分为5组,分别灌服黄蘗、知母、生地黄、女贞子及水。每天剂量:黄蘗与知母为2克生药/公斤,生地黄与女贞子为20克生药/公斤,水为1毫升/鼠。服药2周期内,睾丸切除型大白鼠服4种滋阴药各组的平均收缩压均较服药前1周降低11毫米汞柱以上。黄蘗与知母降低收缩压18与15毫米汞柱;若按降压面积百分比计算,则分别为-18及-16%,与同型对照组比较,降压非常显著(P<0.01),可认为有效。其他各型服不同药的各组,血压降低均在8毫米汞柱以下,其降压面积百分比与对照且比较,降压均不显著。不同的滋阴药,对于各种模型的降压效果各有不同,且对于同一模型的效果也不等。

     

    Abstract: Hypertensive disease is usually treated with "ying tonics" in Chinese medicine. In this paper the hypotensive action of 4 "ying tonics" were studied: Phellodendron sachalinense (Ps), Anemarrhena asphodeloides (Aa), Rehmannia glutinosa (Rg), and Ligustrum lucidum (LI). Rats were made hypertensive by (Ⅰ) burning of left adrenal cortex, (Ⅱ) orchiectomy, (Ⅲ) s.c. injection of testosterone propionate 5 mg/day to female rats for 16 days, and (Ⅳ) s.c. injection of stilbestrol 1—2 mg/day to male rats for 16 days. Blood pressure was measured by the tail plethysmographic method. Starting from the 6th week (but 5th week for model Ⅱ), the "ying tonics" were fed to the rats for 2 weeks. The daily dosages in terms of crude drugs were: 2 g/kg for Ps and Aa, 20 g/kg for Rg and LI, and 1 ml/rat for water as control. During the medication period, 4 "ying tonics" in model Ⅱ caused a drop of systolic pressure of more than 11 mm Hg. The systolic depression for Ps and Aa, moreover, amounted to 18 and 15mmHg resp., or -18% and -16% of hypotensive areas resp., which were highly significant. Thus Ps and Aa were deemed to be effective. Other models showed a depression of blood pressure all below 8 mm Hg. The % of hypotensive areas were non-significant. It is concluded that "ying tonics" did not show similar hypotensive effects in different models, nor even in the same model.
