INHIBITORY EFFECTS OF THREE Gn COMPOUNDS ON TNFα PRODUTION BY MURINE PERITONEAL MACROPHAGES

Abstract
AIM To investigate the effects of isorhapotigenin (Gn-1), resveratrol (Gn-2) and stilbene oligopolymer (Gn-3) on TNFα production by murine peritoneal macrophages stimulated with LPS or rhIL-8. METHODS Inhibition was used to show the inhibitory effect of three compounds on TNFα production by murine peritoneal macrophages measured with crystal violet staining assay of L929 cells. RESULTS Isorhapotigenin(10-5~10-4 mol.L-1), resveratrol(10-5~10-4 mol.L-1) and stilbene oligopolymer(10-6~10-4 mol.L-1) were shown to inhibit TNFα production by murine peritoneal macrophages stimulated with LPS in a dose-dependent manner, IC50 are 4.9×10-5 mol.L-1,5.2×10-5 mol.L-1 and 3.6×10-5 mol.L-1 respectively. Isorhapotigenin(5×10-6~10-4 mol.L-1) and stilbene oligopolymer(10-6~10-4 mol.L-1) were also found to dose-dependently inhibit TNFα production by murine peritoneal macrophages stimulated with rhIL-8, their IC50 are 9.3×10-5 mol.L-1 and 4.6×10-6 mol.L-1 respectively. CONCLUSION The results suggest that the three Gn compounds can inhibit TNFα production by murine peritoneal macrophages, which is probably one of the mechanisms of the anti-inflammatory effects of isorhapotigenin, resveratrol and stilbene oligopolymer.
