DETERMINATION OF FOUR NEO-CLERODANE DITERPENOIDS OF TEUCRIUM SPECIES BY HPTLC SCANNING

Abstract
AIM: To establish a TLC scanning method for the separation and determination of the four neo-clerodane diterpenoids: teucvidin (Ⅰ), 19-acetyl-teuspinin (Ⅱ), teuscorodonin (Ⅲ) and teupernin A (Ⅳ) in T.quadrifarium and the acetone extract of T.pernyi from Teucrium species. METHODS: The sample was extracted with supersonic waves with acetone. Then XAD-2 macroporous resin was used to remove the pigment. EtOAc — n-hexane (6∶5) was used as developing solvent on the HPTLC silica gel plate. After that, the spots were identified by Ehrlich reagent and then determined with a Shimadzu CS-930 TLC scanner. RESULTS: The calibration curves were linear in the range of 0.63~2.18 μg for the four neo-clerodane diterpenoids and the recoveries were 92.4%~97.8%. The method had been applied to the analysis of two plants. CONCLUSION: The method is simple, feasible, rapid and reproducible. It offered a scientific basis for seeking and utilizing botanic resources, and evaluating the quality of herb medicines.
