ANALYSIS OF DL-15-METHYL PROSTAGLANDIN F2α METHYL ESTER

Abstract
This paper deals with the quantitative determination of dl-15-methylprostaglandin F2a methyl ester by thin layer chromatography and subsequent reaction with HCl-EtOH solution. The method has been applied to synthetic samples. The proposed procedure is as follows:Weigh accurately 1~3 mg of sample into a 2 ml bottle, add 2 ml of EtOH to dissolve Spot 10~30 μg of the prepared sample solution on a silica gel G plate, develope with a mixture of dichloromethane—acetone—toluene—cyelohexane(4:6:0.2: 3), locate the spots by iodine vapor and scrape into 10 ml glass stoppered test tubes Add accurately 5.0 ml of 1.5 N HCI-EtOH solution, warm the mixture at 30℃ for 1.5 h, cool to room temperature, centrifuge and take the supernatant liquid for spectrophotometric determination at 239 nm against a reagent blank.The method is simple and rapid. The recovery of dl-15-methyl PGF2a methyl ester is 98.3%.
