EFFECT OF ANTITUMOR ANTIBIOTIC C1027 ON THE EXPRESSION OF GENES RELATED TO APOPTOSIS IN HCT-8 HUMAN COLON CANCER CELLS

Abstract
AIM: To study the effcet of C1027 on the expression of genes related to apoptosis in HCT-8 human colon cancer cells. METHODS: HCT-8 human colon cancer cells were treated with C1027(10 nmol·L-1) for 8 hours. The effect of C1027 on the expression of genes in HCT-8 cells were detected by cDNA arrays, Northern blot and RT-PCR. RESULTS: Hybridization of the entire cDNA populations to Atlas Arrays showed that C1027 up regulated the expression levels of TRAF3, DR4, DR5, MCH4, MCH6, TRIP, Apo-3, ABLL and STAT1 and down regulated the expression levels of rhoC. These changes were confirmed by Northern blot and RT-PCR. CONCLUSION: Most of the genes are involved in TNF receptor family related signal transduction. The results indicate that C1027 may induce apoptosis by regulating TNF receptor family related signal transduction.
