CHROMATOGRAPHIC ANALYSIS OF HEMATOPORPHYRIN DERIVATIVE AND ITS ACTIVE FRACTIONS

The active fractions of hematoporphyrin derivative (HA-308) were prepared by using Bio-Gel filtration procedure. The properties of an active fraction and hematoporphyrin derivative (HPD)were compared with those of photofrin Ⅱ and U. S. made HPD. The result showed that their behaviors on silica gel G thin layer chromatogram and Bio-Gel P-10 column were very similar. The HPLC analysis revealed that our HPD and fraction A all contained the active component corresponding to DHE in photofrin Ⅱ. The examined samples all contained hematoporphyrin and 8(3)-(1-hydroxyethyl)-3(8)-vinyldeuteroporphyrin, but the protoporphyrin was only present in both HPD specimens by TLC analysis.
