HYPOTENSIVE EFFECT OF DIMETHYL TRILOBINE IODIDE (DTI)

  • The hypotensive effect of dimethyl trilobine iodide (DTI) was studied in various anesthesized animals. In dogs, intravenous injection of DTI at the dosage of 0.05~0.1 mg/kg reduced the blood pressure by 42.3~61.5% which lasted about an hour. In cats, the blood pressure was reduced 26.5~43.5% by DTI 0.00625~0.0125mg/kg, the effect persisted 2~4 hours. When DTI 0. 005~0.01 mg/kg was administered to rabbits, the blood pressure was reduced 24.3~61.0%, which lasted more than 5 hours. When given to rats, DTI also decreased the blood pressure markedly. The mechanism of the hypotensive effect of DTI was believed to be related to its ganglionic blockade action.The acute oral and iv LD50 in mice were found to be 522.0 mg/kg and 2.23 mg/kg, respectively.
