PHARMACOKINETIC STUDIES ON DAURICINE

Dauricine is a phenolic alkaloid isolated from Menispermum dauricum D C. In animal experiments dauricine showed an efficient hypotensive action. The pharmacokinetic of dauricine was studied in rabbits. A method was developed for the isolation of dauricine from biological specimens and its quantitative determination was done by Al₂O₃ G TLC and CS-910 Densitometry.The results showed that the logarithm of drug concentration in plasma versus time curve after intravenous administration was fitted to a two-compartment open model with first order elimination. The phamarcokinetic paramenters are as follow:t_1/2α 0.1708 h, t_1/2β 6.506 h, V_c 8.56 L/kg, V_d 46.09 L/kg, K₂₁ 0.9257 h^-1, K₁₀ 0.5834 h^-1, TBCI 81.92 ml/min.
