EFFECTS OF CALCIMYCIN ON PLATELET ADHESION TD CULTURED BOVINE CEREBRAL MICROVASCULAR ENDOTHELIAL CELLS AND ITS ANTAGONISM BY DRUGS

Abstract
Calcimycin induced rabbit platelet adhesion to cultured bovine cerebral microvascu-lar endothelial cells(CMEC)and its inhibition by triazelodiazepine(WEB 2086),1,5-bis-(3,4-dimethoxyphenyl)-tetrahydro-(4H)-pyran(DMPP)and tetrandrine(Tet )were investigated.TheresuIts showed that calcimycin significantly increased platelet adhesion to CMEC,The plateiet adhesionto CMEC was increased by 17.1%:VS controf after stimulation with calcimycin0.01μmol·L-1 for25 min, WEB 2086 0.1,1.0and l0.0μmol·L-1 or DMPP 0.1,1. 0 and l0.0μmol·L-1or Tet0.1,1. 0 and l0.0μmol·L-1 inhibited the calcimycin induced platelet adhesion to CMEC by 9.0,22.9 and 23.1%or9.7, 15.6 and 22.1%or 7.8, 15.6 and 2.6%, respectively.This indicatesthat DMPP and Tet may have perspectives in the prevention af1d treatment of cerebral vascular diseases.
