HPLC DETERMINATION OF DANSHENSU AND PROTOCATECHUIC ALDEHYDE IN RAT PLASMA

  • AIM: To investigate the absorption of Danshensu (Dhpl) and protocatechuic aldehyde (Pal) after im injection of Salvia miltiorrhiza and ig Danshen extract. METHODS: An HPLC method was developed for determination of Dhpl and Pal in rat plasma by internal standard method. In this system, the mobile phase consists of acetonitrile and 0.01 mol.L-1 KH2PO4 (8∶92, adjusted to pH 2.8 with phosphoric acid). The flow rate was 1.0 mL.min-1, the UV detector was set at 280 nm. 4-Hydroxy-benzoic acid was used as the internal standard. The concentration of Dhpl and Pal in plasma was determined by the new method after im Salvia miltiorrhiza injection and ig Danshen extract. RESULTS: The result showed that Dhpl and Pal can be absorbed into blood after injection and oral administration. CONCLUSION: Dhpl and Pal can be used as the index components of the preparations containing Danshen.
